5-Hydroxy L-Tryptophan

Contraindications:

1. Patients on MAOIs (approved 2 weeks after discontinuation)

2. Patients with carcinoid tumors

3. Should not be used concurrently with medications such as: SSRIs, tricyclic antidepressants, MAOIs.

4. Should not be used concurrently with serotonin 5-HT1 receptor agonists including naratriptan, sumatriptan, and zolmitriptan.

5. Patients with ischemic heart disease (Hx of myocardial infarction, angina pectoris, documented silent ischemia)

6. Patients with Hx of coronary artery spasm

7. Uncontrolled hypertension

8. Pregnant women and nursing mothers.

Keep out of the reach of children.

Side effects: Due to excessive serotonin production could include:Nausea, diarrhea, loss of appetite, vomiting, difficult breathing, dilation of pupils, exaggerated reflexes, loss of muscle coordination, blurring of vision and cardiac dysrythmias.

Note: Carbidopa may increase 5-HTP transport across the blood brainbarrier due to decreased peripheral 5-HTP metabolism.

Methyldopa inhibits conversion of 5-HTP to serotonin.

Extracts of Griffonia simplicifolia have been used traditionally in African medicine to inhibit diarrhea, vomiting and constipation, and as an aphrodisiac.

5-HTP is a precursor of serotonin (5-HT).

The diverse physiological funtions of serotonin in the body include actions as a neurotransmitter, a regulator of smooth muscle function in the cardiovascular and gastrointestinal system, and a regulator of platelet function.

Serotonin is involved in numerous central nervous system actions such as regulating mood, sleep and appetite.

In the gastrointestinal system, serotonin stimulates gastric motility.

Serotonin also stimulates platelet aggregation.

As a precursor of serotonin, 5-HTP helps to normalize serotonin activity in the body.

Considerable research has been conducted regarding the activity of 5-HTP. Some of the clinical studies are summarized below:

MOOD - Dysregulation of serotonin metabolism in the central nervous system has been shown to affect mood.

5-HTP helps to normalize serotonin levels and, thereby positively affect mood.

In a double-blind study using objective assessments of mood, researchers in Zurich reported significant improvements in mood with 5-HTP.

1 Likewise, in a double-blind, multi-center study in Germany, researchers reported significant improvements in both objective and self-assessment indices of mood.

2 SLEEP - Many studies have shown that depletion of serotonin results in insomnia, which is reversed by administration of 5-HTP.

Likewise, Soulairac and Lambinet3 reported that 100mg of 5-HTP resulted in significant improvement for people who complained of trouble sleeping.

Furthermore, serotonin is metabolized to the hormone melatonin, which is known to help regulate the sleep cycle, by increasing serotonin levels with 5-HTP, melatonin levels are also increased.

APPETITE - Food intake is thought to suppress appetite through the production of serotonin from the amino acid tryptophan.

Because it is an intermediary in the conversion process of tryptophan to serotonin, 5-HTP may reduce appetite in a similar manner as food intake, but without the calories.

In a recent double-blind, placebo-controlled study, subjects taking 5-HTP lost significant weight compared to control subjects. A reduction in carbohydrate intake and early satiety were seen in the 5-HTP group.

5-HTP leads to an increase in brain serotonin concentration

5-HTP, also known as 5-hydroxytryptophan, is the direct precursor to the neurotransmitter serotonin.

5-HTP works by increasing the amount of serotonin manufactured in the brain.

Although systematically administered serotonin does not readily enter the brain, its direct precursor, 5-HTP does penetrate the blood-brain barrier, and therefore, dietary consumption of 5-HTP leads to an increase in brain serotonin concentration.

Vitamin B-6 is added as it affects both physical and mental health.

It helps with convulsions, headaches, anorexia, depression and numbness, as well as other disorders.

Calcium Citrate is added to help against deficiencies which can attribute to insomnia, muscle cramps,nervousness, convulsions and depression.

Magnesium Citrate is added to assist in calcium uptake, as well as combat deficiencies that can attribute to interference with the transmission of nerve and muscle impulses, depression, dizziness, and muscle weakness and twitching.

Hops may help with anxiety, insomnia, nervousness and pain.

Valerian Root acts as a sedative, as well as help with anxiety, fatigue, insomnia, muscle cramps, nervousness, pain and spasms.

Serotonin is an important initiator of sleep.

In numerous studies 5-HTP has been shown to decrease the time required to get to sleep and to decrease the number of awakening.

It also increases REM sleep and deep sleep.

By increasing the brain's concentration of serotonin, an antidepressant effect is created.

Low levels of serotonin are associated with sleep problems, depression, anxiety, compulsive eating disorders, restless leg syndrome, migraines, fibromyalgia and low pain threshold.

Many people that are afflicted with one of those conditions often have others, which has led some researchers to label them collectively as "Low Serotonin Syndrome."

5-Hydroxytryptophan

5-Hydroxytryptophan (5-HTP), also known as oxitriptan (INN), is a naturally-occurring amino acid and chemical precursor as well as metabolic intermediate in the biosynthesis of the neurotransmitters serotonin and melatonin from tryptophan.

5-HTP is sold over-the-counter in the United States and Canada as a dietary supplement for use as an antidepressant, appetite suppressant, and sleep aid, and is also marketed in many European countries for the indication of major depression under trade names like Cincofarm, LevothymLevotonineOxyfanTelesolTript-OH, and Triptum. Several double-blind placebo-controlled clinical trials have demonstrated the effectiveness of 5-HTP in the treatment of depression, though the quality of the studies has been disputed.

As a therapeutic supplement

Though 5-HTP is not found in food in any significant quantities, it is a chemical involved intermediately in the metabolism of tryptophan, an amino acid found in turkey, milk, potatoes, pumpkin, and various greens. It is often sold as an over-the-counter therapeutic supplement. In this case, it is usually sourced from the seeds of Griffonia simplicifolia. 5-HTP in supplement form is typically sold in 50 mg or 100 mg gelatin or vegetarian capsules.

Research

5-HTP has been studied and shown to be of benefit in the following conditions: primary fibromyalgia syndrome, Friedreich's ataxia,depression, anxiety, binge eating associated with obesity, and insomnia. There is no statistically significant difference between 5-HTP and placebo in treating chronic headaches (primary or otherwise). *
