Whey protein is a mixture of globular proteins isolated from whey, the liquid material created as a by-product of cheese production. Some preclinical studies in rodents have suggested that whey protein may possess anti-inflammatory or anti-cancer properties; however, human data is lacking. The effects of whey protein on human health are of great interest and are currently being investigated as a way of reducing disease risk, as well as a possible supplementary treatment for several diseases.
Whey protein is commonly marketed and ingested as a dietary supplement, and various health claims have been attributed to it in the alternative medicine community. Although whey proteins are responsible for some milk allergies, the major allergens in milk are the caseins
Production
Main article: Whey
Whey is left over when milk coagulates and contains everything that is soluble from milk. It is a 5% solution of lactose in water, with some minerals and lactalbumin. It is removed after cheese is processed. The fat is removed and then is processed for human foods. Processing can be by simple drying, or the protein content can be increased by removing lipids and other non-protein materials. For example, spray drying after membrane filtration separates the proteins from whey.
Whey protein can be denatured by heat. High heat (such as the sustained high temperatures above 72 °C associated with the pasteurization process) denatures whey proteins. While native whey protein does not aggregate upon renneting or acidification of milk, denaturing the whey protein triggers hydrophobic interactions with other proteins, and the formation of a protein gel. Heat-denatured whey can still cause allergies in some people.
Composition
Whey protein is the collection of globular proteins isolated from whey, a by-product of cheese manufactured from cow's milk. Cow's milk is 20% whey protein and 80% casein protein, whereas human milk is 60% whey and 40% casein. Whey protein is typically a mixture of beta-lactoglobulin (~65%), alpha-lactalbumin (~25%), and serum albumin (~8%), which are soluble in their native forms, independent of pH. The protein fraction in whey (approximately 10% of the total dry solids within whey) comprises four major protein fractions and six minor protein fractions. The major protein fractions in whey are beta-lactoglobulin, alpha-lactalbumin, bovine serum albumin and immunoglobulins.
Major forms
Whey protein typically comes in three major forms: concentrate, isolate, and hydrolysate.
- Concentrates contain a low level of fat and cholesterol but, in general, have higher levels of bioactive compounds, and carbohydrates in the form of lactose — they are 29%–89% protein by weight.
- Isolates are processed to remove the fat, and lactose, but are usually lower in bioactivated compounds as well — they are 90%+ protein by weight. Both of these types are mild to slightly milky in taste.
- Hydrolysates are predigested, partially hydrolyzed whey proteins that, as a consequence, are more easily absorbed, but their cost is generally higher. Highly-hydrolysed whey may be less allergenic than other forms of whey. They are very bitter in taste.Function
Whey is essential in the bodybuilding world today because of its ability to be digested very rapidly. This allows the protein to become available for muscle building very quickly. Most commonly it is used after workouts to help increase levels of amino acids in the blood, which are taken up by the muscles to ultimately increase mass. In addition, during exercise, whey helps open up blood flow by inhibiting an angiotensin-converting enzyme which originally constricts blood vessels; this allows better flow of nutrients to necessary areas to help repair and rebuild muscle tissues.
Health effects
The use of whey protein as a source of amino acids and its effect on reducing the risks of diseases such as heart disease and cancer is the focus of ongoing research. Whey is an abundant source of branched-chain amino acids (BCAAs) which are used to fuel working muscles and stimulate protein synthesis. In particular, leucine plays a key role in initiating the transcription pathway that fires up protein synthesis. When leucine is ingested in high amounts, such as with whey protein supplementation, there is greater stimulation of protein synthesis, which may speed recovery and adaptation to stress (exercise).
Whey protein contains the amino acid cysteine which can be used to make glutathione. However, this amino acid is not essential for the synthesis of glutathione and some studies have suggested that the amount of cysteine in the diet may have little effect on glutathione synthesis. However, another study suggested that large amounts of whey protein can increase cellular glutathione levels. Glutathione is an antioxidant that defends the body against free radical damage and some toxins, and studies in animals have suggested that milk proteins might reduce the risk of cancer.
Consumer Reports testing found that due to contamination, some bodybuilding protein shakes purchased online, and in the New York metro area, exceeded USP standards for exposure to heavy metals when three servings a day were consumed. Depending on each product's individual serving-size recommendations, some three-serving samples involved as much as 210 grams (7.4 oz) of powder being tested.

Protein
Bodybuilders often take a powdered form of protein, which contains the essential building blocks for muscle. The powder is mixed with water and often flavoring, resulting in a form marketed as a "shake" (as in milkshake) or "pudding". Protein powder is generally consumed immediately before and after exercising, or in place of a meal. The theory behind this supplementation is that having a sufficient protein intake allows for efficient growth and repair of muscle tissue.
- Whey protein contains high levels of all the essential amino acids and branched-chain amino acids. It also has the highest content of the amino acid cysteine, which aids in the biosynthesis of glutathione. For Bodybuilders whey protein provides amino acid used to aid in muscle recovery.
- Casein protein (or milk protein) has glutamine, and casomorphin.
- Soy protein Soybeans contain isoflavones, a type of phytoestrogen.
- Egg-white protein is a lactose- and dairy-free protein.
- Hemp seed contains complete and highly-digestible protein and hemp oil is high in essential fatty acids.
Some health experts have criticized protein shakes as being unnecessary for most people that consume them, since most users already get enough protein in the normal diet. The body can only metabolize 5-9 grams of protein per hour; excessive daily intake can cause weight gain, kidney problems, or diarrhea.

Branched-chain amino acids
Amino acids are the building blocks of protein; the body breaks consumed protein into amino acids in the stomach and intestines. There are three branched-chain amino acids (BCAAs): leucine, isoleucine, and valine. Each has numerous benefits on various biological processes in the body. Unlike other amino acids, BCAAs are metabolised in the muscle and have an anabolic/anti-catabolic effect on it.
Glutamine
Glutamine is the most abundant amino acid found in human muscle and is supplemented because supplement manufacturers claim the body's natural glutamine levels are depleted during anaerobic exercise. It is argued that bodybuilders should supplement with glutamine, as deficiency may lead to a weakened immune system and wasting of muscle tissue. It is sold as a micronized, instantly soluble powder. Some studies have shown there to be no significant effect of glutamine on bench press strength, knee-extension torque or lean muscle mass when compared to controls taking a placebo, though another study found that glutamine is beneficial in raising T-helper/suppressor cell ratio in long distance runners.
During intense exercise the body produces lactic acid for energy. The lactate is used up for energy and the hydrogen is left, dropping the pH in the muscle, causing a ‘burning’ fatigue. One of the amino acids that your body uses to counteract this acidosis caused by the lactic acid is the glutamine in your bloodstream. In order to replenish the loss of glutamine in the bloodstream, the body catabolises glutamine from the muscle. So supplementing with glutamine ensures there is always a constant supply for the muscles.

Essential fatty acids are necessary nutrients, especially while bodybuilding. Bodybuilders often go on such a low fat diet that they become fat deficient. Fatty fish include salmon, trout, or mackerel; fish oils can also be taken in supplement form. Fish fats cannot readily be made in the body, but are required for various functions within the body to take place.
Polyunsaturated vegetable oils such as corn, sunflower, and safflower oils, cannot provide linoleic acid. Soybean oil is the only supermarket oil that contains linolenic acid. Flaxseed oil is an ideal source of α-Linolenic acid, which can also be found in walnuts and pumpkin seeds.
Obtained from coconut oil, medium chain triglycerides (MCTs) have an unjustified reputation in the world of bodybuilding. It is commonly believed that MCTs cannot be deposited into fat cells, but research has shown this to be incorrect. Although MCTs are rapidly available to the bloodstream, they don't give an athlete more strength, size speed, or endurance.
These are the most benign of fats in that they do not affect cholesterol or prostaglandins (regulators of hormone action) such as some polyunsaturated fats. Monounsaturated fats are found in olive oil and macadamia nuts.
Various supplements in health food stores contain essential fatty acids derived from fish oils and other sources.
Meal replacement products
Meal replacement products (MRPs) are either pre-packaged powdered drink mixes or edible bars designed to replace a prepared meal. Both are consumed in the place of a whole-food meal. Generally MRPs are high in protein, low in fat, have a low to moderate amount of carbohydrates, and contain a wide array of vitamins and minerals.
The majority of MRPs use whey protein, calcium caseinate or micellar casein, soy protein, and egg albumin as the protein source. Carbohydrates are typically derived from maltodextrin, oat fiber, brown rice, or wheat flour. Some also contain flax oil powder as a source of essential fatty acids.
MRPs can also contain other ingredients that are deemed beneficial to bodybuilders. These can include, but are not limited to: creatine monohydrate, glutamine peptides, L-glutamine, calcium alpha-ketoglutarate, additional amino acids, lactoferrin, conjugated linoleic acid, and medium chain triglycerides.
Prohormones
Prohormones are precursors to hormones and were most typically sold to bodybuilders as a precursor to the natural hormone testosterone. This conversion requires naturally occurring enzymes in the body. Side effects are not uncommon, as prohormones can also convert further into DHT and estrogen. To deal with this, many supplements also have aromatase inhibitors and DHT blockers such as chrysin and 4-androstene-3,6,17-trione. To date most prohormone products have not been thoroughly studied, and the health effects of prolonged use are unknown. Although initially available over the counter, in 2004 their purchase was made illegal without a prescription in the US, as it remains so in almost all countries and is proscribed by most sporting bodies.

Creatine
Creatine is an organic acid naturally occurring in the body that supplies energy to muscle cells for short bursts of energy (such as lifting weights) via creatine phosphate replenishment of ATP. A number of scientific studies have shown that creatine can increase strength, energy, muscle mass, and reduce recovery time. In addition, recent studies have also shown that creatine improves brain function and reduces mental fatigue. Unlike steroids or other performance-enhancing drugs, creatine can be found naturally in many common foods such as herring, tuna, salmon, and beef, making it difficult to ban it from sporting competitions.
It increases what is known as cell volumization by drawing water into muscle cells, making them larger. This intracellularretention should not be confused with the common myth that creatine causes bloating (or intercellular water retention). Creatine is sold in a variety of forms, including creatine monohydrate and creatine ethyl ester, amongst others. Though all types of creatine are sold for the same purposes, there are subtle differences between them, such as price and necessary dosage. Non-supplemental suppliers of creatine include various types of offal, red meat, and kidney meat.
In response to claims that creatine can be stressful to the kidneys (due to primary renal elimination via creatinine), creatine proponents cite studies by universities and independent organizations showing no adverse effects.
In The New Encyclopedia of Modern Bodybuilding, 2nd ed., author Arnold Schwarzenegger states:
Creatine monohydrate is regarded as a necessity by most bodybuilders. Creatine monohydrate is the most cost-effective dietary supplement in terms of muscle size and strength gains. … There is no preferred creatine supplement, but it is believed that creatine works best when it is consumed with simple carbohydrates. This can be accomplished by mixing powdered creatine with grape juice.
Thermogenic products
A thermogenic is a broad term for any supplement that the manufacturer claims will cause thermogenesis, resulting in an increased metabolic rate, increased body temperature and consequently an increased rate in the burning of body fat. Until recently almost every product found in this supplement category comprised the "ECA stack": ephedrine, caffeine and aspirin. However, on February 6, 2004 the Food and Drug Administration (FDA) banned the sale of ephedra and its alkaloid, ephedrine, for use in weight loss formulas. Several manufacturers replaced the ephedra component of the "ECA" stack with bitter orange or citrus aurantium (containing synephrine) instead of the ephedrine. To date the effectiveness of this new combination is not conclusive.
Testosterone boosters
There are several naturally occurring plants and vitamins as well as synthetic chemicals that supplement companies claim may produce an increase in testosterone levels. However, the validity of many of these products is questionable due to a lack of valid scientific research showing their effectiveness, and even scientific evidence showing a lack of effectiveness. Some commonly taken supplements of this type are ZMA and Tribulus terrestris.
In a placebo-controlled study, published in the British Journal of Sports Medicine, a statistically significant increase in strength and arm circumference was achieved with an extract of Eurycoma longifolia .
A 2008 study concluded that Eurycoma longifolia is capable of suppressing estrogens which are potent inhibitors of testosterone production.
The effect of Eurycoma longifolia extract on muscle growth was also studied in the animal model. 200, 400, and 800 mg of extract per kg of body weight were fed to male rats for 12 weeks. Then the size of a single muscle, the levator ani was measured and expressed in mg per 100 gram of body weight. At the highest dosage, levator ani size had increased by about 20 percent when compared to controls.
The relatively new drug 4-androstene-3,6,17-trione may increase the testosterone-estrogen ratio.
Excess testosterone can cause undesirable side effects, such as hair loss and acne, and may be converted into estrogens, which can have undesirable effects on males, such as gynecomastia and testicular atrophy.
